The Role of Interferon- in the Activatioin of Tumor-Infiltrating Lymphocytes (TIL) and Lymphokine Activated Killer (LAK) Cells with Interleukin-2 (IL-2)

Abstract
To clarify the role of interferon-¥ã(IFN-¥ã) in the activation of tumor-infiltrating lymphocytes(TIL) and lymphokine activated killer (LAK) cells with interleukin-2 (IL-2), the change of the cvtotoxicity and expression of adhesion molecules were
observed in TIL and LAK cells activated with or without IFN-¥ã.
TIL and tumor cells were isolated from the tumor mass of patients with stomach cancer and peripheral blood lymphocytes (PBL) were isolated from peripheral blood of the same patient.
The isolated TIL and PBL were cultured with IL-2 alone or IL-2 plus IFN-¥ãfor 13 days. The cytotoxicity against autologous tumor cells or cell-line (K562, HeLa) were tested by 4 hr 51Cr release assay.
The TIL cultured with IL-2 plus IFN-¥ãshowed higher cytotoxicity against both autologous tumor cells and cell-line, compared to those cultured with IL-2 alone (p<0.05). IFN-¥ã added LAK cells had higher cytotoxicity against only autologous tumor
compared to those induced by IL-2 alone (p<0.05).
Expression of major histocompatibility complex (MHC) class II molecules and leukocyte functional antigen-1 (LFA-1) on TIL and LAK cells were increased after activation with IL-2 alone or IL-2 plus IFN-¥ãincreased expression of MHC class II
molecules and
inter-cellular adhesion molecule-1 (ICAM-1) on HeLa cells, but did not increased those on K562
In conclusion, IFN-¥ãenhanced cytotoxicity of TIL and LAK cells against autologous and allogeneic tumor cells. It may be attributable to the changes of the expression of MHC class II molecules or adhesion molecules on TIL, LAK cells and tumor
cells.
